I would like to use my 09 giant defy to do some cyclocross races this year. I've never done any cyclocross and really am not concerned with doing well, just wanna have fun and extend my season.
I've got 25's on the bike now. How much thicker can I go? My understanding is that cyclocross bikes usually have 32's?
Anything else I should change other than the tires?

look, measure

see where the tightest clearance is (may be brake bridge, or stays, or fork), and see how much space there is beyond the 25mm tire. Then of course you have to try to figure out the actual (as opposed to labeled) size of prospective tire choices.
