My wheels are Mavic Kysrium, about 4 years old. I want to replace my 23 mm rear Michelin Pro tire with a 25 mm Michelin Pro 2 on bought on sale last year. Will this affect handling or riding safety? I'm a clydesdale, so I'm not worried about weight or absolute performance, just safety.

No. Lots of people run a larger tire on the rear. No problem. Doesn't even have to be the same model or brand.
