I am running King hubs (which have Shimano compatible hub shells) and a Campy 10 speed drivetrain. With repsect to cassette choices, is American Classic the only company that makes a compatible cassette?

Anyone tried running a Shimano cassette in a Campy 10-speed drive train?

Believe so. You could get a JTEK Shiftmate, and then you could use a Shimano 10 cassette.

If you search JTEK on RBR you should find threads talkin' about the Shiftmate.

Several companies make Campy 10spd spaced cassettes that fit on Shimano freehubs. In addition to American Classic, Wheels Manufacturing makes the Accelerator 10 cassette, and Cycle Dynamics makes an all titanium Campy-on-Shimano 10spd conversion cassette.
